The identification of specific cell markers and characteristics of stem cells has been only partially successful thus far. Several specific cell molecules are currently in use for stem cell isolation, but there is no agreement on unique markers and “stemness signatures”. For example, bone marrow transplants have succeeded using stem cells selected by “specific markers” which function is poorly characterized and none are distinctive. The SME developed an alternative way to select stem cells: selection by function. Cell death (apoptosis) plays a pivotal role in the regulation of cell growth in the fetus and adult. The resistance to death is an intrinsic mechanism in a variety of stem cells and disappears upon differentiation and proliferation. Through intensive laboratory work, the SME formulated a procedure that exploits this resistant characteristic of stem cells so that they survive treatment while other cells undergo apoptosis. The procedure involves binding of cell surface receptors that normally signal cell death in differentiating and proliferating cells, thereby providing the means for isolation of stem cells from non-stem cells. Since stemness is basically a state in which some cells can get into and out from, using the functional characteristics of a stem cells makes much more sense when trying to isolate these populations.

This phenomenon is reversible upon recovery from the chronic inflammation. • Platform for testing neutralizing agents for their capacity to restore the expression of the T cell biomarker and enhanced immune competence. • The T cell biomarker expression may be used as a diagnostic/prognostic tool to detect chronic inflammation‐induced immunosuppressive environment. Thus therapeutic effectiveness of the drugs can be enhanced and side effects can be decreased. The invention involves glucuronide ligand conjugation to magnetic based nanoparticles and then labeling with I-125 and I-131, separately SEM analyses showed that the particles size are 40-60 nm intervals. Labeling yields are found to be 89.4% for I-131 labeled uracil-o-glucuronid conjugated silanated magnetic particles, 92.6% for I-131 labeled uracil-n-glucuronid conjugated silanated magnetic particles. While cellular incorporations for n and o-glucuronides are high comparing to uracil, incorporations for urasil, n and o-glucuronide conjugated magnetic particles are always high. Cellular incorporations for ligand conjugated magnetic particles are much higher under magnetic field.

The QLISA technology is a new in vitro diagnostic test for highly sensitive detection of inflammatory biomarkers and differentiation of IBD vs. IBS. QLISA uses quantum dots to measure inflammatory biomarkers and proprietary surface modifications to provide a superior alternative to the ELISA technique by requiring much smaller sample volumes and simpler assay protocol while preserving ELISA’s sensitivity. The platform is applicable to a 96 well plate format or flat slides of your material of choice. These unique features translate into significant practical advantages of using QLISA as a platform for in vitro diagnostics applications. The initial application of QLISA being developed at Drexel is a diagnostic test discriminating between IBS (irritable bowel syndrome) and IBD (irritable bowel disease). As many as 4 million people worldwide (1 million Americans) suffer from a form of IBD and more than 15 million have IBS, with estimated medical costs in the USA at over $2 billion/yr when adjusted for loss of productivity. IBD refers to two chronic diseases of intestinal inflammation: ulcerative colitis and Crohn's disease. Diagnosing these diseases is an expensive and lengthy process requiring knowledge of medical history, blood tests, X-rays, endoscopy, tissue histopathology and testing of stool samples. As a result, early differentiation of IBS vs. IBD is difficult, which leads to unnecessary excessive testing of IBS patients and significant delays in much needed treatment of IBD patients. Thus, there is a critical need to develop a rapid, low cost, reliable diagnostic test to differentiate common intestinal symptoms of IBS from IBD, to monitor remission and relapses of IBD, and to evaluate success of or need to change medication. The QLISA technology circumvents issues related to sampling and is envisioned for use in a clinical lab environment as a platform for a broad variety of in vitro diagnostic (IVD) tests, including IBD vs. IBS. The proposed product is a medical device for evaluation of the depth of anesthesia awareness during surgery. The device is based on functional near-infrared (fNIR) technology and relies on optical techniques derived from the physical principles of light absorption and reflectance to detect changes in the hemodynamic response of the cortex to brain activation. A monitoring apparatus for objectively and continuously quantifying the depth of anesthesia of a patient during surgery would not only reduce the incidence of patient awareness (0.13%), but also would allow the anesthesiologist to administer the minimal dose of anesthetic required to achieve the desired depth of anesthesia. The proposed system is intended to be utilized in any settings where sedation is given. As many as 95 million patients annually in the United States undergo procedures for which anesthesia depth monitoring would prove useful, thus affording a potentially large market. The fNIR Depth of Anesthesia Monitoring Technology is available for licensing from Drexel University now.

The resulting product will be a box where a subject will insert their hands; the box will turn on automatically for a few seconds and turn off with an audible and visual alert that hands are now disinfected and can be removed. The system will be designed to work with all standard disinfectants.

Wound Clinics who see patients with challenging wounds need faster evaluation of the healing process to save costs, improve patient outcomes and to be able to transfer some of the wound care to less specialized environments. Yearly incidence of skin ulcers (pressure, venous and diabetic) in the U.S. is estimated to be 6 million patients, and the annual costs for management of these wounds are greater than $20 billion. The current paradigm is measurement of wound size, by tracing or by digital imaging. This superficial evaluation delays proper treatment and results in continuation of non-efficacious products/methods which ultimately lead to amputations. The proposed device uses NIR technology to measure the level of oxygenated and deoxygenated hemoglobin at the wound site and compares it to a control/non-wound site. The time course of the oxygenated hemoglobin was found to be a strong indicator of wound healing. One PCT patent application and one provisional patent application to be converted into a PCT this November protect the method and the system design architecture. We estimate that shortening by 50% the observation time based on wound size,the cost of care for chronic wounds would be reduced by 30-60%. Third-party payers are reimbursing chronic wound care. The business plan of this project just won the 2008-2009 Wharton Business Plan Competition. The Wallace H. Coulter Foundation has invested $200,000 in the research effort during 2006 and 2007 through a competitive proposal process and University City Science Center invested $100,000 in 2009 through its QED program. Scientists have recently discovered that by delivering electrical stimulation to specific regions of the brain (epileptic focus), they are able to prevent the occurrence of seizures. This technique, also known as neuromodulation, is currently used in open-loop systems where uncontrolled electrical currents are delivered to the nervous system constantly. Due to fear of adverse effects, scientists have begun to focus their efforts on developing a more sophisticated closed-loop system that predicts the occurrence of seizures and delivers the stimulation only when necessary. Current methods of closed-loop neuromodulation are developed based on prediction using EEG recordings,which is difficult and have so far only proven to be 50% effective in predicting seizures. In comparison, we aim to develop an implantable closed-loop neuromodulation device that uses single-neuron recordings to predict seizures. Single-neuron recording is a cutting edge technology in neurological signal recordings. Our Ceramic Based Multi-Site Electrode, developed by Dr. Karen Anne Moxon, Professor of Biomedical Engineering, Neurobiology and Anatomy at Drexel University, allows us to record accurately and chronically from multiple single-neurons in the brain, a feat which has so far been unattainable. This breakthrough in material science and neurological recording allows us to develop an Epilepsy Control System, an implantable device the size of a quarter that can be chronically implanted into an epilepsy patient’s neural tissues using current available surgical methods.

The objective of this project is to develop a new staple for small lumen structures fabricated from an FDA approved biocompatible, biodegradable polymer that delivers bioactive agents at a pre-programmed rate to improve healing. The Drug Delivery Surgical Staple is a rapid and efficient anastomotic platform comprised of a staple, shaped like a push clip, which would be sutured by the head to the bile duct. Then, the tail of the staple would be fed into an incision in the small intestine where it would mate with the washer to connect the two tissues together. The staple was designed with threads on its outer diameter in order to lock it with the washer. This way, the staple could snap into the washer and allow for some variation in tightness between the tissues. After time, the staple and washer would slowly degrade in the body as the tissues heal, with the washer releasing wound healing agents, chemotherapeutic agents and/or radioactive therapy thereby assuring delivery of treatment to the site where it is needed most, the surgical margin.

Ultrasound examination via natural orifices, such as transesophageal ultrasound for echo cardiography and endovaginal ultrasound for diagnosing gynecologic disorders, are gaining increasing popularity. However,training physicians and medical students to perform these procedures is difficult and expensive due to the complex anatomy of the orifices, shortage of volunteer subjects, and very limited capabilities of available but expensive human mannequin-based systems. To fill this gap in the medical education market Drexel’s clinicians and engineers have developed VEUSim, a virtual reality training system with haptic interface to provide real-life experience of ultrasound examination via a natural orifice. VEUSim has unlimited capabilities for modeling various anatomical structures and creating and modifying associated pathologies, and a unique trainee/instructor interface for recording and quantitative evaluation of trainees’ performance and progress. VEUSim has been developed as a Virtual Endovaginal Ultrasound Simulator (hence the name, VEUSim) as described below, and can be easily adapted to simulate ultrasound examination via any other natural orifice such as esophagus, colon etc. In future this same technology can potentially be utilized for training other Ultrasound, surgery and tracheotomy techniques.
